Carboxyfluorescein diacetate succinimidyl ester (CFSE) is a fluorescent dye which can track the cell division. In Vitro: Carboxyfluorescein diacetate succinimidyl ester (CFSE) is a fluorescent dye which can track the cell division.To examine the ability of immune cells to migrate into live slices, a single-cell suspension of autologous splenocytes or PBMC is isolated, label them with CFSE, and add them to the top of the slices on day 2 of culture. After 6 additional days of culture, the CFSE-labeled immune cells (green) are found to have migrated throughout the slices. No positive cells are found in the slices without the addition of CFSE-labeled immune cells. The presence of adoptively transfer leukocytes in the slice cultures 6 d after addition to the culture system demonstrates the feasibility of using slice cultures as a preclinical model for the evaluation of adoptive cellular immunotherapies.
Protocol:Cell Assay: Single-cell suspensions of splenocytes (or PBMC) are stained with 1 µM CFSE in PBS for 9 min at 37°C, combine with 20 mL of 10% FBS RPMI-1640 medium at RT for 2 min, centrifuge, wash, and count. 2×106 CFSE-labeled splenocytes are added to the top of the slices in 24-well membrane culture insert. After 6 d co-culture, slices are fixed with 4% paraformaldehyde and stained the nuclei with DAPI. Finally, slices are visualized with the confocal microscope with Z stack of 2 µM/step. The image process is analyzed by software.
